Abstract

Jamu, an Indonesian traditional herbal medicine has been used traditionally by the local community to keep their fitness or prevent some illness. Scientific examinationand efficacy study of Jamu are growing in these recent years after the announcing ofJamu scientification program by the government of Republic of Indonesia. In order to provide scientific evidences of some Indonesian plant used for Jamu andvegetable, this study is aimed to analyze the active components of the leaf of Carica papaya Linn. (papaya), one of the well known and widely used plants for Jamu and vegetable. Furthermore, biological activities of the compounds including the peroxynitrite-scavenging and anti-obesity effects and their content will be evaluated simultaneously together with the validation and standardization of the quantification method using a High Performance Liquid Chromatography (HPLC). Therefore, the active principle of C. papaya, including its content and biological activity can berevealed and understood. This study was designed, because there has been no report on the quantification of the active compounds of C. papaya leaf. Although several studies have reported the biological activity of extract of leaves of C. papaya, investigation on the main active constituent of C. papaya leaf and its biological activities is still lacking. In the first year research, a total of seven flavonoids including of quercetin 3-(2Grhamnosylrutinoside), kaempferol 3-(2G-rhamnosylrutinoside), quercetin 3-rutinoside, myricetin 3-rhamnoside, kaempferol 3-rutinoside, quercetin, and kaempferol have been isolated and elucidated from the leaf of C. papaya. In this second year research, establishing of the simultaneous quantification method of the identified compounds using HPLC technique has been performed along with evaluation of their peroxynitrite-scavenging activities. The content of kaempferol 3-(2Grhamnosylrutinoside) was significantly higher than other identified compounds (123.18 mg/g BuOH fraction and 7.23 mg/g MeOH extract). All of the substances exhibited potent activities on peroxynitrite scavenging (IC50 ≤ 4.15 μM); which were stronger than the positive control, L-penicillamine (6.90 μM). The results of the present study demonstrate the potent antioxidant flavonoids of C. papaya leaf, with kaempferol 3-(2Grhamnosylrutinoside) as the major one. The manuscript containing these research data has been accepted in Asian Pacific Journal of Tropical Biomedicine (a Scopus Q3- indexed journal) and now under proofreading prior to the publishing
